A Microfluidic Static Droplet System for Antimicrobial Susceptibility Test

In clinical research, antimicrobial susceptibility tests (ASTs) are performed to check antibiotic sensitivity of pathogens. It is an important clinical step to prevent the prescription of ineffective antibiotics that may affect the resistance of pathogens towards subsequent antibiotic treatment. For this, rapid determination of antimicrobial susceptibility is essential. However present method take days with limited point of care accessibility. Towards this end, we worked on developing an integrated microfluidic system capable of performing AST studies within few hours. In present research, after testing different microfluidic approaches, an easy to use droplet based Static Droplet Array (SDA) was proposed to perform AST test on E. coli bacteria against ampicillin. SDA comprises of an array of sixty droplets of 30 nL volume with no cross contamination. Owing to its small size, the device has the potential to be used as a point of care diagnostic applications. The result shows that the device takes 2-7 hours depending on the cell density of the bacterial culture (105-108 CFU/mL). The number of cells inside the nanolitre droplets ranges from 20-100. The main highlights of this device is its easy loading with just one pipetting step and the scant volume of culture (2µL) it requires.
