Differences of Metabolic Perturbations between Thai Jasmine Rice and its Brown Planthopper (BPH)-resistant Isogenic Lines During BPH Infestation using 1H NMR and GC-MS Approaches

Brown planthopper (BPH; Nilaparvata lugens) is a phloem feeding insect which is one of the most serious threats to rice crops in many countries throughout Asia. Little is known about the metabolic responses of Thai Jasmine rice (KD) and its BPH-resistant rice isogenic lines (IL7 and IL308) to BPH attack. Polar metabolome and fatty acids from the leaf extracts during 8 days of the infestation were investigated by 1H NMR spectroscopy and GC-MS, respectively. Physiological changes of the rice isogenic lines were different based on the quantitative trait loci of BPH resistance. Multivariate models were capable of distinguishing metabolic profiles between the susceptible and the resistant rice varieties throughout the infestation. Metabolic pathway analysis suggested that BPH infestation has caused the metabolic perturbations in transamination, amino acid metabolism, shikimate, purine/pyrimidine, fatty acid and gluconeogenesis pathways. From metabolomic differences, the susceptible and the resistant rice varieties seemed to employ different pathways to fight against BPH infestation. Several potential metabolic pathways for acclimatization and defense mechanisms against BPH infestation have been identified. These findings provide a valuable insight into BPH resistance mechanisms in Thai Jasmine rice.
