Single-Cell Dissection of Gene Transcription Regulation Networks that Control Cell Cycle Decisions and Cell Fates after DNA Damage

The faithful replication of DNA during each cell division is essential, because uncorrected DNA errors can severely disturb cell function and ultimately lead to cancer. In response to DNA damage mammalian cells have a variety of mechanisms to either repair DNA, enter alternative cell cycle fates, or undergo programmed cell death. Individual cell fate thus depends on the decision which of these mechanisms is activated. Using a combination of single-cell RNA-sequencing and live cell imaging, we are now able to investigate in unprecedented detail how cells coordinate the transcriptional response to DNA damage. We begin to understand which gene regulation networks determine cell fates and therapy resistance of cancer cells.
