Abstract



Extraction of Multiple Pure Biomolecules using a Microfluidic System

Extraction of biomolecules like DNAs or proteins from separation gel are commonly realized with selective gel slicing, followed by electroelution, or gel dissolution and centrifugation based extraction [1-3], etc. However, most methods are time-consuming. Here we report a disposable microfluidc device with embedded extraction chamber for rapid separation and extraction of multiple biomolecule fragments by a portable in-house built detection system. Isotachophoresis followed by gel electrophoresis was used to preconcentrate and separate biomolecule fragments on chip. Multiple target biomolecule fragments were transferred into respective extraction chambers by open/close onchip microvalves. Transferring was started and stopped by monitoring software with trigger function. The optimized working conditions and results obtained on the application of developed microfluidic system for extraction of multiple biomolecule fragments will be presented and discussed.
