Modeling and Imaging the Immune Response in 3D Cell-based Assays

The immune system plays an important role in disease progression for a variety of indications. Still, the vast majority of in-vitro cell-based assays neglects the immune system. The complexity of sourcing matched donor material, differentiating between different cell types in assay readouts and handling the dynamics of these systems imposes difficult hurdles in implementation.
We present two examples of human in-vitro 3D microtissue assays including PBMCs to model the immune response in oncology and T1 diabetes applications. Live-cell, high-content imaging is used as primary readout to assess the dynamics of the immune response. Image analysis of the 3D microtissues allows for the generation of robust quantified data on immune cell penetration and target-cell viability.
