Renal Regenerative Potential of Different Extracellular Vesicle Populations Derived from BM-MSCs

Extra-cellular vesicles (EVs) derived from human bone marrow mesenchymal stromal cells (MSCs) promote regeneration in different animal models of AKI, in a manner comparable with the cells of origin. We know that EVs isolated from cultured MSCs are heterogeneous in terms of size and density. It is not clear which population of the EVs is responsible for the pro-regenerative effect. To clarify the nature of EV population that is able to induce renal regeneration, we evaluated the effect of different EV-populations, separated by differential ultracentrifugation on AKI recovery and characterized their molecular content (mRNA, miRNA and proteins). Moreover, we tested in vivo and characterized EVs derived from different stem cells/progenitor sources in particular, EVs derived from resident renal and liver stem/progenitor cells. We correlated the different molecular components of different EV populations with their capacity to improve renal regeneration.
