Abstract

Making Chemically Intelligent Choices for HPLC Method Development

K K Bhagchandani, Senior Director, Advanced Chemistry Development Inc

Many times, during HPLC Method Development, we have seen people trying to optimize their methods and are trying to expand the design space. While this is a required exercise, however, if we make logical and chemically intelligent choices from the start of during early stages, this will be so much easier to achieve this. What is important is that QbD is recommending us to build the quality inside the method rather than trying to enhance it at a later stage. This short presentation will highlight how the available tools from ACD/Labs can help users to make better choices in the beginning of the method development to expect better success of the method. Some of the assistance it can provide is about 1.Which of our standard method is more likely to work? 2.What type of pH-range we need to choose? 3.What will be a good combination of solvents etc ? 4.Which of the trial methods are better and should be pursued further? 5.Which of the analytes is eluting at what retention time? 6.Is there any change in the elution order by changing the conditions/parameters of the methods?
