Dimiter Dimitrov's Biography



Dimiter Dimitrov, Head/Senior Investigator, National Institutes of Health

Dr. Dimitrov graduated and completed his PhD at the University of Sofia, Sofia, Bulgaria, in 1976. Thereafter he worked in the Bulgarian Academy of Sciences where he defended his ScD in 1985 and was Professor of Biophysics until he joined the National Cancer Institute (NCI) of the National Institutes of Health (NIH), USA, in 1990. He was tenured as Senior Investigator in 1993 and appointed at the Senior Biomedical Research Service in 2008. His research group includes molecular biologists who are experts in display/screening/libraries methodologies, antibody engineering, and protein biochemistry, structural biologist and computational scientist. His major long-term goal is the development of clinically useful therapeutics and vaccines based on human monoclonal antibodies including engineered antibody domains. He has authored and coauthored more than 300 articles cited more than 10,000 times, and is inventor or coinventor of more than 50 inventions, patent applications or patents. This Keynote Talk will give an overview about the current state of the ADC field and will discuss in more details site-specific conjugation which has certain advantages compared to random conjugation.  A number of methods for site-specific conjugation will be discussed including engineering sites for conjugations, using naturally occurring sites for conjugation and enzymatic vs chemical methods for generation of site-specific ADCs. Conjugation of antibodies with small molecules through carbohydrates will be discussed in more details and latest results will be presented including generation of ADCs against cancer-related targets.
