My lab is interested in elucidating how dynamic methylation of histone proteins within nucleosomes contributes to the regulation of transcription. In particular, we are investigating the function and regulation of Jumonji C domain-containing histone demethylases. We are approaching these challenges by combining chemical synthesis, protein biochemistry, mechanistic enzymology and cellular studies. In addition, my lab is interested in the catalysis and evolution of substrate specificity of a pair of related and mechanistically novel bacterial enzymes that methylate the peptidyltransferase center of the ribosome, causing resistance to five chemically distinct classes of antibiotics.
