Expanding Reproducibility and Coverage for Biofilm Metabolomics Workshop

Biofilms continue to be a major source of morbidity and mortality, espectially in the field of orthopaedics. Indeed they are now the main source of infection in Gram-positive bacteria. With increasing incidence of MRSA and VRSA strains in the hospital environment, new methods are required to investigate pathogenesis and biofilm dispersal.
We have developed methods for high throughput extraction of metabolites from both biofilms and planktonic cells, providing a large number of samples per experiment, so that multiple biological replicates of multiple strains under multiple conditions can be assayed. This leads to large datasets where bioinformatic approaches must be applied to deal with instrument drift and batch-to-batch variation. In addition, we have developed methods to expand the detection of both primary and secondary metabolites in multiple systems. We present data combining these methodologies from a drug mode-of-action study on a clincally obtained strain of staphylococcus.
