Facile synthesis of universal DNA nanoparticles with rolling circle amplification for cancer diagnosis

DNA has been applied to numerous biomedical fields with the development of nucleic acid-based nanotechnologies. As a novel material for target detection, DNA has been used to fabricate a barcode system with a various structures. In this work, multifunctional DNA nanoparticle by rolling circle amplification (RCA) is synthesized for target detection with diverse functional moieties. DNA nanoparticle barcode system was designed to present fluorescence with nucleic acid staining dyes or quantum dots and express functional biomolecules. The tumor-specific antibodies can be coated on DNA nanoparticle surfaces through biotin-streptavidin interaction. As a proof of concept, the capability for target detection of multifunctional DNA nanoparticle barcode system was demonstrated by direct cell detection as a simple method. The functionalized DNA nanoparticle suggests a new route for the simple and selective recognition of cancer cells.
