Epigenetically modified bone marrow derived mesenchymal stem cells improved cardiac function after myocardial infarction in rats

Bone marrow mesenchymal stem cells (BMMSCs) have generated a great deal of interest in the regenerative medicine and tissue engineering. In the present study, we isolated mesenchymal stem cells from rat bone marrow and cardiomyocytes from neonatal rats characterized them on the basis of the presence of surface markers by Immunocytochemistry. We have treated mesenchymal stem cells with demethylating agents, which has the ability to transdifferentiate mesenchymal stem cells into cardiac cells. Co-culture of normal and treated mesenchymal stem cells with cardiomyocytes was done after labeling them separately and their ability to fuse with each other was analyzed at different time points by fluorescence microscopy and flow cytometric analysis. The treated MSCs were also transplanted to the rat model of myocardial infarction and cardiac functional studies were performed by echocardiography. Mesenchymal stem cells expressed surface markers while cardiac specific proteins were present in the isolated cardiomyocytes. The fusion of the two types of cells increased exponentially. Our study demonstrated study that the treated mesenchymal stem cells have the ability to fuse with the cardiomyocytes more as compare to untreated. These stem cells have potential to differentiate into cardiomyocytes and therefore can be used as a cellular therapy for cardiovascular disorders.




Adipose Derived Stem cells in Facial Rejjuvenation Surgery

Since their discovery 2 decades ago, Adipose derived Stemcells(ADSC) has become the mostly investigated adult stem cells with ready clinical applications. They are easily accessible and plentiful compared to bone marrow derived stemcells, with ratio of 20:1. ADSC is pluripotent, multi-lineage and posses high degree of regenerative and anti-inflammatory potentials. Because these qualities ADSC is deemed to be an ideal cell/tissue product in plastic and reconstructive surgery for tissue grafting , volume replacement, repair and regeneration especially in the sub dermal fibro-vascular tissue of face that that has undergone age related deterioration. Complying with all the US FDA protocols and regulations we have performed a series of ADSC grafting during various facial plastic surgery procedures from 2001 --2015 and our results are promising. However more long term investigations are needed to fully understand and optimize the results and for future advancement The technical aspects and protocols of harvesting the ADSC form different donor sites, separation, processing , purification and engraftment of the cells are described in detail.
