Single Cell Analysis for HCS
Thierry Dorval, Group Leader, Institut de Recherches Servier
Robustness of phenotypic approaches relies mostly on the physiological relevance of the cellular models used within screening campaigns. In this context, and thanks to the recent advances in cell cultures, cellular models are now displaying an increasing level of heterogeneity. This heterogeneity is turning obsolete regular data analysis approaches where each readout per experiment can be captured by averaging the population. Not only the heterogeneity is a phenotypic signature by itself, but it also provides a powerful way to characterize and understand mechanisms of action. However the amount and complexity of generated data requires specific tools and analysis. Through a series of case studies, we will present practical ways of automatically extracting information from those multiparametric single cell data, and show how those data can also influence quality controls. We will focus our attention on practical aspects of the analysis, and how it is currently deploy in the Servier’s drug discovery pipeline.
