Therapeutic antibody discovery requires a diverse toolkit of technologies to enable early discovery through to manufacture and quality control/lot-release of final product. This technology snapshot presentation will introduce a range of technologies from Promega applicable to early stage discovery and characterisation of antibodies including fusion protein approaches for oriented antigen display, magnetic bead platforms for antibody purification and ‘on bead’ conjugation of antibodies for the preparation of antibody drug conjugates (ADC’s) and conjugation of antibodies to pH sensitive dyes for internalisation studies. A range of functional cell-based bioassays will also be discussed including reporter gene assays for the assessment of ADCC and ADCP mechanism of action, T-activation cell lines for development and characterisationof bispecific antibodies and novel assays which recapitulate the immune synapse for the study of immune checkpoint targets such as PD1/PDL1 and CTLA-4.
