Quantitative high-throughput phenotypic screening platform to identify antibiotic-persister cells

We developed a high-throughput microfluidic-microscopy platform to screen transposon-induced mutants of Mycobacterium smegmatis (M. smegmatis) for those within aberrant persistence phenotypes to the anti-tuberculosis drug isoniazid (INH) in order to identify genes responsible for this phenotype. Using this platform we screened 1100 strains and identified 11 novel mutants that show substantially increased or decreased tolerance to INH. These results yield new insight into the field that may provide new targets for pharmaceutical development.
