Imaging and Functional Manipulation in Cells and Cell Mimics

Our lab is pursuing three parallel areas of investigation to image and manipulate model membranes and cells. (1) We use DNA-lipid conjugates to create complex membrane architectures to study specific membrane-based transformations, in particular membrane fusion. These conjugates can be used to transfer molecules to and through cell membranes, e.g. for imaging in dense tissue. (2) We use imaging mass spectrometry using a NanoSIMS to provide high spatial resolution quantitative information on the composition of membrane domains in model membranes with a lateral resolution approaching 50 nm. This can be applied to the analysis of the lateral organization of model membrane-cell membrane interfaces. (3) We use a variety of split GFPs to manipulate interactions within and between cells using light. This genetically encoded system can be used to modulate specific targets in cells or to deliver components to cell surfaces. My talk will be some combination of these three seemingly disparate approaches.
