ADMET Optimisation in Drug Discovery

ADMET optimization is typically done in all phases of discovery to select optimal ADME properties of drug candidates and reduce failure rate, as development of any drug amounts to almost a billion dollars nowadays. Starting with in silico models a screening phase is usually performed generating a multitude of ADMET data characterizing a drug candidate’s properties. The final stage in drug discovery is the profiling phase where drug candidates are investigated including prediction of human PK and dose prediction. The interface between Discovery and Development is discussed as well. Examples of each phase are given and specific challenges surrounding the following points are discussed and commented on: Applicability domain and validity, decision points, preclinical strategies and false positive rates.
