Usually they populate at the hot spots of binding in protein surface-exposed binding pockets. Therefore, their crystallographic characterization is usually successful and the obtained structures provide a valid starting point for a subsequent optimization by growing the initially discovered fragments in the binding pocket via the decoration with suitable side chains. |
