As we age, our cells undergo a process where they enter a senescent state as a potent tumor suppression mechanism, but this cellular change may also contribute to the aging pathology seen within tissues including inflammation and bioenergetic compromise. In this talk the use of single cell analysis is used to dissect the cellular transition from a quiescent to senescent state.

NanoVelcro-Embedded Chips that Enable Molecular Analysis of Single Circulating Tumor Cells

This presentation will introduce a new category of NanoVelcro Chips capable of not only capturing circulating tumor cells (CTCs) with high efficiency, but also enabling highly specific isolation of single CTCs immobilized on the embedded nanosubstrate without contamination by white blood cells (WBCs).  The individually isolated CTCs can then be subjected to mutation analysis by next generation sequencing, providing insights to molecular mechanisms responsible for the malignant transformation in cancer.

Clonal Analysis of Cancer at the Single Cell Level

Nearly all solid tumors consist of heterogeneous mixtures of genetically abnormal and genomically unstable cells.  As such, the basic unit of the disease is the single cell.  The goal of our work is to enable the molecular dissection of cancer at the single cell level and to extract the maximum useful information in the most cost efficient manner.  We will discuss methods for rapid CNV analysis and focal resequencing for SNV as applied to circulating tumor cells and core biopsies.
